The X chromosome DNA test determines the likelihood that two sisters have the same biological father.
Since the two sisters share the same biological mother, then this one must participate in the DNA test so that the laboratory can discard the X chromosome coming from the mother and thus concentrate the research on the one coming from the father.
If the mother cannot participate in the X chromosome test then you must do a sibling test.
